Functional single-cell assays with droplet flow cytometry

In this session, our Field Application Scientist presents a novel workflow combining Samplix Xdrop with the BD FACSDiscover S8, leveraging CellView imaging to capture both population-level data and high-resolution views of individual droplets.
This approach allows:
- Visualization of double-emulsion droplets
- Detection of cell encapsulation within droplets
- Direct observation of cell–cell interactions, such as NK cell-mediated killing of target cells
- Measurement of functional activity, including Granzyme B substrate signal within single droplets
By compartmentalizing cells in droplets, signals remain isolated, avoiding the mixing effects seen in bulk assays. This opens new possibilities to:
- Study immune cell function at true single-cell resolution
- Monitor cytotoxicity, cytokine capture, or antibody secretion in confined microenvironments
- Strengthen workflows for cell and gene therapy development, where functional validation is critical
